To analyze the expression of endothelin receptor subtypes ETA ETB ; in the heart, binding assays were performed in the presence or absence of the subtype-specific endothelin receptor ligands BQ 123 3 mol L ; and or BQ 3020 3 mol L ; . The assay buffer for binding studies contained 1 mg mL bacitracin, 100 mmol L TrisHCl, 5 mmol L MgCl2, and 1 g L BSA, pH 7.4, in a total volume of 150 L. The 125IET-1 tracer concentration was kept constant at 40 000 counts per minute per tube while the concentration of unlabeled ET-1 was increased from 0 to 25 nmol L competition studies with "cold saturation" ; . Samples from crude plasma membranes were used at a concentration of 0.53 mg protein mL 1 according to Hocher et al.15 Binding studies were performed at room temperature for 120 minutes. Nonspecific binding was assessed in the presence of excess ET-1 5 mol L ; . After adding 1 mL of cold binding buffer, free and receptor-bound radioactivity was separated by centrifugation at 30 000g 4C ; for 15 minutes, and the pellets thus obtained were washed 2 additional times with 1 mL of cold binding buffer. 125I was counted in a Packard gamma counter 78% counting efficiency for 125I ; . Protein concentrations were determined according to Lowry et al.14 Bmax and Kd values were calculated by linear regression analysis of Scatchard plots. About Mylan. In order to establish conditions required for optimal killing, B-CLL cells were initially exposed to increasing doses of CYC202 for 4, 8, 16, and 72 hours and cellular viability and apoptosis assessed at each time point Figure 1 ; . Treatment with 1 g ml had little effect on induction of apoptosis and cell viability in any of the samples, whereas 2.5 g ml affected most but not all of the samples. In contrast, within 24 hours of the start of treatment with CYC202 at 5 g ml, there was a dramatic induction of apoptosis accompanied by loss of viability and by 48 hours of treatment most B-CLL cells were undergoing apoptosis. Little dose-dependency was observed with concentrations above 5 g ml. We found that the first signs of apoptosis were detectable between 4 and 8 hours of treatment with 5 g ml CYC202 and the proportion of viable cells greatly diminished by 16 hours data not shown and camptosar. Eling, and function. Treatment with wortmannin was confirmed to prevent the increase in Akt activity observed in AT1AKO hearts 1 wk post-MI Fig. 5A ; . It was also found to restore the decreased apoptosis in granulation tissue cells Fig. 5B ; . Importantly, treatment with wortmannin partially but significantly reversed the attenuated post-MI remodeling and dysfunction at the chronic stage Fig. 5C ; . Overall, these findings suggest a significant involvement of increased activity of Akt signaling in post-MI benefits properly observed in AT1AKO mice.
20 000 years ago, at the height of the last glacial maximum on Earth, the climate in Siberia was harsh--and yet there was no ice! Pretty strange. However, a new climate model blames dust from the deserts of Asia. It seems likely that the dust darkened the Siberian snow, causing seasonal snow to melt due to greater absorption of solar radiation and capsicum. AMGEN INC. NOTES TO CONSOLIDATED FINANCIAL STATEMENTS Continued ; repurchase of a portion of the Convertible Notes discussed below ; or 5.77 per note is being accreted to the balance of the Convertible Notes and recognized as interest expense over the life of the Convertible Notes using the effective interest method. The holders of the Convertible Notes had the right to require us to repurchase all or a portion of their notes on March 1, 2005. On March 2, 2005, as a result of certain holders of the Convertible Notes exercising this March 1, 2005 put option, we repurchased .59 billion aggregate principal amount of Convertible Notes for their then-accreted value of , 175 million in cash, or approximately 40% of our then outstanding Convertible Notes. Upon the repurchase of such Convertible Notes, a pro rata portion, million, of the related debt issuance costs were immediately charged to interest expense in the quarter ending March 31, 2005. We made an aggregate cash payment of million to the holders of the Convertible Notes who did not exercise the put option and continued to hold outstanding Convertible Notes subsequent to March 1, 2005. This payment was approximately equal to 1.25% of each Convertible Note's then-accreted value and will be amortized to interest expense over the life of the remaining outstanding Convertible Notes using the effective interest method. Concurrently, we amended the terms of the Convertible Notes to add an additional put date in order to permit the remaining holders, at their option, to cause us to repurchase the Convertible Notes on March 1, 2006 at the then-accreted value. Accordingly, the portion of the Convertible Notes outstanding at December 31, 2004 not repurchased on March 2, 2005 was classified as long-term debt in the accompanying Consolidated Balance Sheet. On May 6, 2005, we exchanged new zero-coupon senior convertible notes the "Modified Convertible Notes" ; and a cash payment of approximately million for approximately 95% of the remaining Convertible Notes then outstanding. In August 2005, we exchanged substantially all of the remaining Convertible Notes. The changes to the Convertible Notes outstanding as a result of the May and August 2005 exchanges combined with those made in March 2005 are being accounted for as a debt modification. Accordingly, all cash paid to the holders of the Modified Convertible Notes and Convertible Notes collectively, the "convertible notes" ; is being amortized to interest expense over the life of the convertible notes using the effective interest method, and the costs incurred to modify the terms of the convertible notes were expensed as incurred. As of December 31, 2005, convertible notes with an accreted value of .8 billion and having an aggregate face amount of .36 billion remained outstanding. Because the holders of substantially all of the convertible notes did not require us to repurchase such notes on March 1, 2006, the next available put date, the convertible notes were classified as non-current in the accompanying Consolidated Balance Sheet as of December 31, 2005. The significant terms of the convertible notes are as follows: Holders of Convertible Notes and Modified Convertible Notes may convert each of their notes based on a conversion rate of 8.8601 shares of common stock of Amgen as defined below. The conversion price per share of the convertible notes as of any day will equal the original issuance price plus the accrued original issue discount to that day, divided by the conversion rate or .15 as of December 31, 2005. The conversion price per share at issuance of the Convertible Notes was .61. While the Convertible Notes are convertible into common stock at any time, the Modified Convertible Notes can only be converted if: 1 ; the closing price of common stock exceeds the conversion price per share during a defined period at the end of the previous calendar quarter, 2 ; we call the Modified Convertible Notes for redemption, or 3 ; we make certain significant distributions to common stockholders or enter into specified types of corporate transactions and carboplatin.
